Abstract/Notes | Genus of Megalocytivirus (family Iridoviridae) have been known as one of the most devastating viral disease in both of marine and freshwater aquaculture in Asia. This research had been conducted to know the disease-agent caused mass mortalities in sea bass, Lates calcarifer, and its pathogenecity to humpback grouper, Cromileptes altivelis. Fish samples were obtained from private net cage at Pegametan Bay Singaraja, Bali when mass mortality was occurred. The samples were processed for laboratory analysis. The isolate was recognized as SBIV by PCR analysis using primer derived from RSIV-DNA (red sea bream iridovirus-DNA) sequence. Histopathological observation revealed necrotic and degenerating cells on the spleen, kidney and liver of sea bass. Pathogenecity test comprising injection of spleen, kidney, and liver inoculum resulted in 100% mortality within 7 days, while pathogenecity test by cohabitation among sea bass and humpback grouper resulted in 80%mortality within 9 days. PCR analysis indicated that moribund and dead humpback groupers infected with SBIV. Histopathological observation revealed enlarged cells as well as cells in the spleen, kidney and liver. Otherwise, the control group was unaffected and showed normal visceral organs. This result concluded that mass mortality of sea bass was caused by SBIV,that pathogen to humpback grouper.
